Biomotif and Karolinska Institutet have co-pioneered the pI-Trap Åcelus system for automated, MS-compatible, high speed, isoelectric, pre-fractionation of peptides & proteins. The pI-TRAP Åcelus system automatically pre-fractionates a complex protein sample into 30 fractions in about one hour to radically leverage biomarker discovery work-flows. Biomotif AB is also proud to have partnered with Spark Holland BV, a world-class provider of proven analytical technologies, to develop the auto-sampling & fraction collection capabilities of the pI-Trap Åcelus.

PRE-FRACTIONATION ENABLES DISCOVERY

Pre-fractionation of complex clinical samples containing 100,000s of proteins (with a vast dynamic range) is key to rigorous biomarker discovery. Protein pre-fractionation before analysis by LC/MS/MS allows researchers to dig deeper into the proteome and reproducibly discover more specific biomarkers.

MS-COMPATIBLE PRE-FRACTIONATION ESI

MS analysis of peptides & proteins occurs in the Liquid-Phase. The pI-Trap Åcelus rapidly fractionates peptides and proteins in solution, and provides an orthogonal separation to hydrophobicity based HPLC.

DISRUPTIVE TECHNOLOGY

At the heart of the pI-Trap Åcelus system is Biomotif’s proprietary multi-junction capillary isoelectric focusing (MJ-CIEF) pI-CELL. After isoelectric focusing, a carrier buffer mobilizes the focused proteins towards the fraction collector. This process can cause band broadening of any residual proteins. The Biomotif pI-CELL uniquely enables recursive re-focusing after every fraction collection step, to counteract any band broadening effects. This unique technology allows up to 30 (re-focused) fractions to be collected automatically, while preserving the advantages of the primary isoelectric separation and all in about one hour.
